Liver stem cells grown in culture: Therapeutic benefit demonstrated

Feb. 25, 2013 ? For decades scientists around the world have attempted to regenerate primary liver cells known as hepatocytes because of their numerous biomedical applications, including hepatitis research, drug metabolism and toxicity studies, as well as transplantation for cirrhosis and other chronic liver conditions. But no lab in the world has been successful in identifying and growing liver stem cells in culture -- using any available technique -- until now.

In the journal Nature, physician-scientists in the Pap? Family Pediatric Research Institute at Oregon Health & Science University Doernbecher Children's Hospital, Portland, Ore., along with investigators at the Hubrecht Institute for Developmental Biology and Stem Cell Research, Utrecht, Netherlands, describe a new method through which they were able to infinitely expand liver stem cells from a mouse in a dish.

"This study raises the hope that the human equivalent of these mouse liver stem cells can be grown in a similar way and efficiently converted into functional liver cells," said Markus Grompe, M.D., study co-author, director of the Pap? Family Pediatric Research Institute at OHSU Doernbecher Children's Hospital; and professor of pediatrics, and molecular and medical genetics in the OHSU School of Medicine.

In a previous Nature study, investigators at the Hubrecht Institute, led by Hans Clever, M.D, Ph.D., were the first to identify stem cells in the small intestine and colon by observing the expression of the adult stem cell marker Lgr5 and growth in response to a growth factor called Wnt. They also hypothesized that the unique expression pattern of Lgr5 could mark stem cells in other adult tissues, including the liver, an organ for which stem cell identification remained elusive.

In the current Nature study, Grompe and colleagues in the Pap? Family Pediatric Research Institute at OHSU Doernbecher used a modified version of the Clever method and discovered that Wnt-induced Lgr5 expression not only marks stem cell production in the liver, but it also defines a class of stem cells that become active when the liver is damaged.

The scientists were able to grow these liver stem cells exponentially in a dish -- an accomplishment never before achieved -- and then transplant them in a specially designed mouse model of liver disease, where they continued to grow and show a modest therapeutic effect.

"We were able to massively expand the liver cells and subsequently convert them to hepatocytes at a modest percentage. Going forward, we will enlist other growth factors and conditions to improve that percentage. Liver stem cell therapy for chronic liver disease in humans is coming," said Grompe.

U.S. Department of Agriculture (USDA) and university scientists have found that cattle temperament influences how animals should be handled, how they perform and how they respond to disease.

The team of researchers looked at stressful eventssuch as weaning, transportation and vaccinationthat beef cattle experience during routine management practices. The researchers examined interrelationships of stress and cattle temperament with transportation, immune challenges and production traits.

Studies were conducted by animal scientist and research leader Jeff Carroll at the Agricultural Research Service (ARS) Livestock Issues Research Unit (LIRU) in Lubbock, Texas; associate research professor Rhonda Vann at Mississippi State University's Brown Loam Branch Experiment Station; animal physiologist Ron Randel at Texas AgriLife Research, The Texas A&M University (TAMU) System, in Overton; and endocrinologist Tom Welsh, Texas A&M AgriLife Research and TAMU Department of Animal Science, in College Station.

Between 24 and 36 calves were used for each study, depending on the trial. An exit velocity system, which measures the rate at which an animal exits a squeeze chute and crosses a certain distance, was used to select for temperament. A pen scoring system was used in conjunction with exit velocity to calculate an overall temperament score for cattle selected as the calmest, the most temperamental or as intermediate.

When challenged with a bacterial toxin, cattle showed dramatic differences in sickness behavior, depending on their temperament. The more temperamental animals failed to show behaviors that allow detection of sick animals, whereas calm animals immediately displayed visual signs and became ill. Studies also revealed that temperamental cattle did not have the same vigorous immunological response to a vaccine as less temperamental cattle in the same herd.

In related research, the team found that the main cause of stress for cattle was not transportation itself, but being handled and loaded into a trailer.

However, transportation duration and conditions were found to have negative effects on intramuscular fat or marbling, which is used for fast sources of energy by cattle being transported. Marbling determines the quality grade of beef. Lower levels of marbling reduce quality grade. Temperamental cattle have less fat stores, indicating that temperament makes a difference in the final quality grade.

Microsoft joins list of companies recently hacked

REDMOND, Wash. (AP) ? Microsoft has joined the list of prominent technology companies confirming they have been hit by a recent computer hacking attack.

In a blog posting Friday, Microsoft said it had found no evidence that any customer data had been heisted.

Microsoft Corp. gave few other details about the break-in, except to say that was it similar to a hacking attack that online social networking leader Facebook Inc. disclosed last week. Facebook had said its investigation had discovered other companies had been hacked, but didn't identify the other victims.

Like Facebook, Microsoft says it is still investigating how malicious software was planted on what it said were a small number of its computers.

Online messaging service Twitter also recently disclosed that hackers may have stolen information about 250,000 of its users.

GOSHEN, Indiana ? Union workers at a northern Indiana trailer-hitch manufacturer have voted to forego arbitration and accept a severance agreement that will pay the most senior employees $36,000.

Most of the 350 United Steelworkers Local 9550 workers at Cequent Performance Products in Goshen supported the package in votes tallied Friday night, Local 9550 Vice President Deb Hathaway said.

"No matter what they give us it's still not enough for us losing our jobs," she said.

Steelworkers Sub District 4 Director Mike O'Brien said 240 of the 350 union members at the plant voted. He didn't disclose vote totals.

"It was certainly not unanimous but it wasn't close," O'Brien said. "I think a lot of people looked at, 'Well, this is what the company is offering and we take a chance going to arbitration.'"

Cequent, a subsidiary of Bloomfield Hills, Michigan-based TriMas Corp., announced in November it would move operations from the 450-employee plant 25 miles southeast of South Bend to Reynosa, Mexico, to lower shipping costs.

Workers with less than a year's seniority will receive $500 while those with 30 years or more will receive $36,000. Workers also will receive company health benefits for at least a month after they are laid off. The total severance package is worth more than $3.5 million, O'Brien said.

Layoffs began Friday, but O'Brien said he didn't know how many workers lost their jobs.

More than half the employees are scheduled to remain on the job through the end of June, with the last due to be let go in December.

Winfrey, 'Beasts' actress honored at Essence lunch

BEVERLY HILLS, Calif. (AP) ? Nine-year-old Oscar nominee Quvenzhane Wallis toted a plush, dog-shaped purse with jeweled handles as she rubbed shoulders with Oprah Winfrey and Alfre Woodard.

The star of best-picture contender "Beasts of the Southern Wild" was among the honorees Thursday at Essence magazine's sixth annual Black Women in Hollywood awards luncheon.

Standing on a step so she could speak at the podium, Quvenzhane thanked God, "Beasts" director Behn Zeitlin and "my baby sitter that was on set."

Quvenzhane is the youngest best-actress nominee in Oscar history and one of only 10 African-Americans ever recognized in the category.

Winfrey was teary-eyed as she took the stage after being introduced by the graduating class of her namesake Leadership Academy for Girls in South Africa.

"I am humbled by this day," she said. "There's nothing better than being honored by your own."

Essence President Michelle Ebanks said the afternoon's honorees "are changing the very definition of power" with their presence in Hollywood.

Woodard said she always recognized herself in the pages of Essence and recalled that when she and actresses C.C.H. Pounder and Loretta Devine first arrived in Hollywood, "they didn't know what to do with (us)."

"We were young and gifted and looking way the hell black," Woodard said. "And we still have our original noses!"

Actress Gabrielle Union moved the audience as she accepted the Fierce & Fearless award. Union said she had long pretended to be fierce and fearless but actually reveled in gossip and "took joy in people's pain and tap-danced on their misery." She said she posed for photos in such a way to "minimize my blackness" and didn't speak out against racism in school or in Hollywood when she was younger.

"Real fearless and fierce women admit mistakes and work to correct them," she said. "We stand up and we use our voices for things other than self-promotion. We don't stand by and let racism and sexism and homophobia run rampant on our watch. Real fierce and fearless women celebrate and compliment other women and we recognize and embrace the notion that their shine in no way diminishes our light, and actually makes our light shine brighter."

Actress Naomie Harris, who plays Eve Moneypenny in "Skyfall," was among those touched by Union's words.

"I'm in the place you were," Harris said as she accepted the Shining Star award. "I feel as though I have traveled thousands of miles to actually listen to that speech, to learn, to be inspired and to be reminded of who I am."

Writer-producer Mara Brock-Akil cried throughout her acceptance speech, saying, "All I ever wanted to do was tell our story."

Other stars attending the luncheon at the Beverly Hills Hotel included Blair Underwood, Laila Ali, Star Jones, Jill Scott and Tracee Ellis Ross.

Grammy-winning singer Miguel performed his winning hit, "Adorn," and rapper-actor Common did an impromptu freestyle during a brief technical difficulty, working the names of the six honorees into a quick rap.

Defence Minister rejects claims that troops not well trained

Defence Minister Dr Jonathan Coleman is rejecting claims the NZDF Crib 20 rotation to Bamyan in Afghanistan, which suffered the loss of five lives, was inadequately trained.

The NZ Herald reported today on a critical evaluation report written during CRIB 20?s pre-deployment training.

?The NZDF prides itself on its professional standards. This rotation was assessed by the NZDF as trained to assume full operational duties in Bamyan,? said Dr Coleman.

?The report to which the NZ Herald refers was one of 23 evaluations conducted during pre-deployment training which contributed to a final training report,? he said.

?There was nothing unusual or different about this process, which has been applied on all rotations to help achieve the highest standards.? I have been assured by the NZDF that any gaps identified were addressed before the CRIB took operational responsibility,? said Dr Coleman.

?The Defence Force lost five soldiers during this deployment in two separate insurgent incidents.? The NZDF is still evaluating what took place and the government is waiting to receive the outcome of their inquiries.?

?It is mischievous for Labour to suggest that funding was a factor in pre-deployment training and incorrect for Labour to say funding has compromised troop safety in Afghanistan. NZ efforts in Afghanistan have been well funded.?

?The Provincial Reconstruction Team in Bamyan is well trained and achieves excellent results in a difficult operating environment. The quality of our Defence Force people is internationally recognised.? said Dr Coleman.
